Heterologous Expression of AmA1 Gene Encoding Storage Protein of Amaranthus in Transgenic Lettuce (Lactuca sativa)

Abstract

|
To transform the lettuce with AmAl gene encoding the seed storage protein of Amaranthus hypochondriacus, AmAl gene was subcloned into plant binary vector with cauliflower mosaic virus (CaMV) 35S promoter and nopaline synthase terminator (3¢¥NOS). The recombinant binary vector was used to transform Lactuca sativa using Agrobacterium-mediated transformation procedure. Transgenic shoots were induced on MS medium added with 0.1 §·¡¤L^(-1) NAA, 0.5 §·¡¤L^(-1) BA, 50 §·¡¤L^(-1) kanamycin, and 300 §·¡¤L^(-1) cefotaxime. Roots from shoot were induced on rooting medium based on MS medium containing 50 §·¡¤L^(-1) kanamycin and 250 §·¡¤L^(-1) cefotaxime. The presence of AmAl gene was confirmed by PCR followed by DNA hybridization in the transgenic plants. The expression of AmAl gene was observed in transgenic plant by RT-PCR method.
